Tris Buffer Formulations

10 mM Tris, 0.1% Sodium Azide, pH 8.0 ± 0.1

This buffer was designed for CEA (Cat. No. C0224), PSA (Cat. Nos. P0723P0724P0725P0715), and Non-Complexing PSA (Cat. No. P5125), but it is also a suitable Tris buffer for use when no additional buffer salts are required. 

Ingredients
Tris Base: 1.210 g/L

Sodium Azide: 1.0 g/L
5M HCl: As needed
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Tris Base and Sodium Azide in approx. 90% of final volume with cold dH2O.
2. Add cold dH2O to obtain final volume.
3. Adjust to pH 8.0 ± 0.1 with 5M HCl or 5M NaOH as needed. Filter as needed.


10 mM Tris, 150 mM Sodium Chloride, 0.1% Sodium Azide, pH 8.0 ± 0.1

This buffer is designed for use with Recombinant Apoferritin. It is also a suitable Tris buffer for use when additional buffer salts are needed. 

Ingredients
Tris HCl: 1.296 g/L

Tris Base: 0.215 g/L
Sodium Chloride: 8.766 g/L
Sodium Azide: 1.0 g/L
5M HCl: As needed
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Tris HCl, Tris Base, Sodium Chloride, and Sodium Azide in approx. 90% of final volume with cold dH2O.
2. Adjust to pH 8.0 ± 0.1 with 5M HCl or 5M NaOH as needed. 
3. Add cold dH2O to obtain final volume.
4. Check pH and adjust to pH 8.0 ± 0.1 with 5M HCl or 5M NaOH as needed.


20 mM Tris, 1 mM EDTA, 50% Glycerol, pH 8.5 ± 0.1 

This buffer was designed for Cardiac Myoglobin (Cat. Nos. M0724M0725), but it is a suitable Tris buffer for use when the additional stability provided by EDTA and glycerol are needed. 

Ingredients
Tris HCl: 1.950 g/L

Tris Base: 0.920 g/L
EDTA, Tetrasodium: 0.416 g/L
Glycerol: 500 ml/L
5M HCl: As needed
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Tris HCl, Tris Base, and EDTA in approx. 45% of final volume with cold dH2O.
2. Adjust to pH 8.5 ± 0.1 with 5M HCl or 5M NaOH as needed. 
3. Add cold dH2O to obtain approx. 50% of final volume.
4. Add cold Glycerol to obtain final volume. Mix thoroughly and chill.
5. Check pH and adjust to pH 8.5 ± 0.1 with 5M HCl or 5M NaOH as needed.


20 mM Tris, 150 mM Sodium Chloride, pH 7.4 ± 0.1 

This buffer was designed for a1 Antichymotrypsin (Cat. No. A1824). This solution is similar to the buffer described above for Recombinant Apoferritin, but it contains a stronger Tris concentration and a slightly lower pH. 

Ingredients
Tris Base: 2.420 g/L

Sodium Chloride: 8.766 g/L
5M HCl: As needed
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Tris Base and Sodium Chloride in approx. 90% of final volume with cold dH2O.
2. Adjust to pH 7.4 ± 0.1 with 5M HCl as needed.
3. Add cold dH2O to obtain final volume and chill.
4. Check pH and adjust to pH 7.4 ± 0.1 with 5M HCl or 5M NaOH as needed.
5. Filter using 0.45 µm filter unit.


20 mM Tris, 280 mM Sodium Chloride, 5 mM Calcium Chloride, 0.1% Sodium Azide, pH 8.0 ± 0.1 

This buffer was designed for our C-Reactive Protein products (Cat. Nos. C0128, C0129), but it is suitable for use when a stronger concentration of Tris and buffer salts, plus calcium chloride, are needed. 

Ingredients
Tris Base: 2.423 g/L

Sodium Chloride: 16.363 g/L
Calcium Chloride: 0.735 g/L
Sodium Azide: 1.0 g/L
5M HCl: Approx. 3 ml/L
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Tris Base and Sodium Chloride in approx. 90% of final volume with cold dH2O.
2. Adjust to pH 8.0 ± 0.1 with 5M HCl as needed.
3. Add cold dH2O to obtain final volume and chill.
4. Check pH ,adjust to pH 8.0 ± 0.1 with 5M HCl or 5M NaOH as needed, then add Sodium Azide.
5. Add Calcium Chloride and allow to dissolve completely. (IMPORTANT: The pH CANNOT be adjusted after adding Calcium Chloride.)
5. Filter using 0.2 µm filter unit.
