Phosphate Buffer Formulations

10 mM Sodium Phosphate, 150 mM Sodium Chloride, 0.05% Sodium Azide, pH 7.2 ± 0.1

This buffer was designed for our preparations of α-Fetoprotein (AFP; Cat. Nos. A0723A0724) and it is a versatile sodium phosphate buffer for general use.

Ingredients
Sodium Phosphate, Dibasic, anhydrous: 1.022 g/L
Sodium Phosphate, Monobasic, monohydrate: 0.386 g/L
Sodium Chloride: 8.766 g/L
Sodium Azide: 0.500 g/L
5M HCl: As needed
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Sodium Phosphate (dibasic and monobasic) and Sodium Chloride in approx. 10% of final volume with dH2O at room temperature.
2. Add cold dH2O to obtain final volume.
3. Check pH and add Sodium Azide.
4. Adjust to pH 7.2 ± 0.1 with 5M HCl or 5M NaOH as needed. Filter as needed.

10 mM Potassium Phosphate, 100 mM Sodium Chloride, 0.1% Sodium Azide, pH 7.6 ± 0.1

This buffer was designed for our partially purified Carcinoembryonic Antigen (Cat. No. C0222). That product has been discontinued, but this is still a suitable potassium phosphate buffer for general use.

Ingredients
Potassium Phosphate, Dibasic, anhydrous: 1.480 g/L
Potassium Phosphate, Monobasic, anhydrous: 0.204 g/L
Sodium Chloride: 5.844 g/L
Sodium Azide: 1.0 g/L
5M HCl: As needed
5M NaOH: As needed
Distilled Water (dH2O)

Directions
1. Dissolve Potassium Phosphate (dibasic and monobasic) and Sodium Chloride in approx. 80% of final volume with dH2O at room temperature.
2. Add cold dH2O to obtain final volume.
3. Check pH and add Sodium Azide.
4. Adjust to pH 7.6 ± 0.1 with 5M HCl or 5M NaOH as needed. Filter as needed.
